Test Code APCPNL Activated Protein C Resistance

Important Note

Acceptable initial test to detect activated protein C resistance due to a factor V Leiden variant.

Additional Codes

30127

Methodology

Electromagnetic Mechanical Clot Detection

Specimen Type

Blood

Preferred Collection Container

Blue

Collection Volume

2.7 mL in 2.7 mL tube

Minimum Collection Volume

2.7 mL in 2.7 mL tube

Collection Instructions

Collect specimen in blue top (citrate) tubes. If the specimen is drawn into a plastic Vacutainer tube, then the tube should be gently inverted no more than three or four times; excessive mixing will activate platelets and affect the test result. Specimen should arrive at lab within 4 hours of collection; transport at room temperature. If unable to test samples within 4 hours, perform transfer instructions.

Centrifuge samples at 1500 g for 15 minutes.  Remove plasma with plastic transfer pipette and place into a plastic tube.  Centrifuge the transferred plasma at 1500 g for 15 minutes.  Transfer plasma to new plastic tubes (1 mL aliquots per tube) and freeze. Store at -20° C for up to 4 weeks. Transport frozen specimen on dry ice.

Rejection Criteria

Specimens that are clotted, hemolyzed, diluted with IV fluid, received unfrozen more than 4 hours after collection, insufficient volume, improperly labelled, not double spun (not platelet poor), thawed specimens, improper or no anticoagulant.

Specimen Storage and Stability

Freeze double spun aliquots within 4 hours after collection.  Samples may be frozen at -20° C for up to 3 months or at -70° C for up to six months.
